Stamp duty at applicable percentage (say 5% or 6%) - depending upon the State in which the Property is located - on the value of the property is to be paid at the time of Registration with the Sub-Registrar's office.

Mere Rs. 100 stamp paper is not enough.

Both the seller (wife) and the buyer (husband) have to be present before the Sub-Registrar's office.  Further consideration (sale price) has to be paid by the husband to the wife.
