x chased y but during the course y fell in a river cause the death of y is it amount to murder

No, here x will not be liable for murdering y because there was no intention of X to kill Y. Here section 302 of the IPC will not apply. However, section 304 of the IPC i.e. "punishment for culpable homicide, not amounting to murder" can be applied.
