Regarding partition of a building

We have 340 sq meter of land which is a joint mutation land of 3 brothers and 1 sister. 3 brothers have constructed jointly 3 flats on ground floor within 326 sq meter, the remaining space is a vacant area and will go to sister. The 3 flats are of unequal dimensions and unequal structure.

 Now we would like to make a partition deed. First i would like to know if partition deed of only the building portion according to present diagram of each flat is possible or not ?

No. the partition deed should be for the entire property.  if it partly remains, then it amounts to conveyance.
