Sir, If We Didn't get the Reply of RTI within 30days, thenbit is necessary to make 1st Appeal or we can Directly Appeal to the State Information Comission??

You should file a first appeal to the First Appellate Authority.

First appeal has to be filed before filing second appeal to state commission.
