The immovable Property must be registered in your name. Transfer of Property Act stipulates that any transfer of property of more than Rs One hundred MUST be registered with the Registrar
So get the sale agreement registered with the registrar of your District. The Sale Deed ( Sale agreement) must be written/typed on Non judicial Stamp Paper of the requisite value

If the property is purchased by you through a registered deed, you are owner, ask the Municipality to inquire and sanction the building plan. If they do not agree approach court.

What is the basis of complaint of your brother and what is the nature of ownership of the property from whom you have purchased the same?
Inform whether the deed of purchase is registered or not.

If you have purchased it by sale deed, that remains and you will be the owner, however it is to be seen whether the vendors had absolute title or ownership to sell this property. Take the advise of a local advocate and proceed as per his further advise.
