No,after having sold the questioned property by your mother to you,your mother mother has lost her ownership right,title and interest in the property so she can not execute will in relation to the said property and even if she makes than such will deed shall be null and void.

Has the mother mentioned in sale deed that only 300 sft. is constructed and not full property?

Being a transaction within your family members your mother can refuse to write a will. Even if it is done you try to amicably sort out with your brother. If not agreed to consult a local lawyer to challenge. Have you applied for transfer of putta and transfer of property in your name in village records. If not you may do the same immediately based on the sale deed.
