if there is no reply for an RTI for more than 40 days,filed with a govt dept in TN, can i move this to Tamil Nadu State Info commission directly or does it still need to go through the first appellate.

Before moving to the State Information Commission you must exaust the remedy available under the Act ,that is the Appeal to the First Appellate Authority.

Yes as properly advised by experts, you may exhaust the remedy available with the first appellate authority and then move to SIC

Before moving to SIC , you should exhaust the remedy available one that is the appeal to the first appellate authority as the every appeal made to the Commission should be accompained by the certified copy of the orders which preferred in earlier appeal.
